Position Focus:

Under the direction of the principal investigator, the Research Associate (RA) will maintain and help build on a high-throughput screening pipeline to identify immunologic antigens; this process involves molecular biology, cell culture, cell staining, and cell sorting. The Research Associate must have in-depth knowledge of molecular biology, genomics, and/or cellular immunology; and must have prior experience with mammalian tissue culture. The Research Associate will be responsible for working with and providing technical expertise to other members of the laboratory. The RA will also be responsible for lab maintenance and management, including but not limited to supply inventory, equipment management, and lab safety protocols. Works with the principal investigator to maintain and build on core techniques in the laboratory involving high-throughput cellular co-culture, cell staining, and cell sorting. The candidate will also be responsible for organizing the lab, acting as point-of-contact for vendors, maintaining equipment, and ordering day-to-day consumables. Trains incoming laboratory personnel (undergraduate, postgraduate, graduate students and postdoctoral associates) in complex experimental procedures and methods, and is the contact person for modifications, updates, and renewals of laboratory IACUC mouse protocol, and EH&S modifications, updates, and renewals. Oversees daily lab operations related to the projects in the laboratory. Conducts research requiring specialized techniques, including: mouse colony management (including genotyping); breeding and tracking of genetically modified mice; the preparation of genetically modified human and mouse cell lines via genome editing; liver subset cells’ isolation, collection and analysis of mouse tissues (with a focus in liver) using molecular, microscopic and related techniques; preparation of samples for analyses via confocal and super resolution microscopy and analysis of gene expression via bulk and single-cell RNA-sequencing; CRISPR screens in cell lines. Bioinformatic skills or willing to learn them for analysis of next generation sequencing data. Train incoming lab personnel in experimental methods developed in our laboratory. Analyzes and interprets scientific results. Presents reports of results at regular laboratory meetings.

Essential Duties

1. Designs, plans, implements and evaluates complex multifaceted research projects/systems, including method development, adaptation and validation. 2. Collaborates with PI to define research endeavors and the development of research hypothesis and approach. 3. Solves complex methodology, protocol, procedural and research problems through design of techniques, procedures and policies that will achieve research goals. 4. Develops long range plans for supplies and equipment to ensure smooth operation of laboratory. 5. Investigates, analyzes and evaluates complex data, data collection systems and methods to reach scientific conclusions and ensures the integrity of research data. 6. Collaborates with PI to coordinate major lab renovations working with Facilities, Health and Safety, and Purchasing to ensure deadlines, safety standards and purchases are according to plans and within budgets. 7. Prepares scientific reports and papers for research proposals and published reports. 8. May perform other duties as assigned.

Required Education and Experience

Master’s Degree in a scientific discipline and three years of experience or an equivalent combination of education and experience.

Required Skill/Ability 1:

Excellent written and verbal communication and organizational skills. Ability to communicate verbally and in writing in a clear, concise manner is necessary. Strong interpersonal skills. Excellent computer skills, including Microsoft Office (Word, Excel, PowerPoint). Excellent customer service skills.

Required Skill/Ability 2:

Strong technical and scientific abilities. Will perform complex laboratory experiments and procedures with guidance from the PI. Laboratory techniques: cell culture and aseptic technique, nucleic acid isolation, PCR, biosafety level 2 work.

Required Skill/Ability 3:

Laboratory techniques: cell culture and aseptic technique, nucleic acid isolation, PCR, biosafety level 2 work. Skilled in mouse genetics, breeding, maintenance, genotyping and tracking of mouse lines, and preparation and analysis of mouse tissues, with a focus in liver research.

Required Skill/Ability 4:

Basic knowledge of mouse husbandry focusing on generating genetically mutant mouse lines, genotyping, and maintaining logs of mouse lines. Demonstrated ability to assemble mouse experimental cohorts and perform mouse procedures, including timed matings and injections.

Required Skill/Ability 5:

Ability to multi-task effectively with competing demands between research projects and lab personnel tasks. Proven ability to develop long term plans for supplies/equipment to insure smooth operation of the lab. Strong attention to detail. Ability to work independently and with others in a team environment. Lead by example. Strong organization and record keeping skills.

Preferred Education, Experience and Skills:

Experience & Molecular and Cancer biology, mammalian tissue culture; DNA, RNA & protein isolation & analysis; Molecular biology techniques: Immunocytochemistry, Fluorescence in situ Hybridization, FACS; Microscopy; Mouse colony maintenance, including genotyping; knowledge in molecular biology, next-generation sequencing technologies & prior experience with lab automation.
