The overall purpose of the work is to develop a human pluripotent stem cell disease model for the rare genetic condition Acrodysostosis characterised by short stature and other skeletal malformations. The post, funded by a grant from the Medical Research Council (UKRI), seeks to understand the underlying molecular pathways which are dysregulated in Acrodysostosis types 1 and 2 and ultimately find ways of correcting them. The post involves maintaining and characterising both induced pluripotent stem cells and human embryonic stem cells with and without disease relevant mutations, differentiation of these cells to skeletal lineages, and assisting with somatic cell reprogramming and CRISPR-Cas9 gene editing. The successful candidate will carry out a range of techniques including PCR, genotyping, flow cytometry, immunofluorescence, as well as histological, immunohistochemical and biochemical assays. The appointee will work with postdoctoral scientists, PhD students and technicians carrying out research using both primary human cells and cell lines in different cell biological contexts. The post is based in the Michael Smith Building of the School of Biological Sciences, Faculty of Biology Medicine and Health, University of Manchester. It will involve working with the members of the Kimber group and their collaborators in other Schools within Manchester and other Universities and Institutes.
