You will work on research to evaluate a PET imaging biomarker for neuropathological changes in the earliest stages of Alzheimer’s disease. Mitochondrial dysfunction appears to be an early consequence of AD pathology. The novel PET radioligand [18F]BCPP binds to mitochondrial complex I to provide a measure of mitochondrial density. Pilot work preceding this study has shown reduction of [18F]BCPP PET ligand binding throughout the brain cortex of people with early AD in association with reduced cerebral blood flow. This study would confirm these observations and extend them to explore ligand binding in people at high risk but without clinical expression of dementia.
The work will be predominantly clinical. There also will be opportunities to perform image analysis and wider in silico molecular and epidemiological analyses related to testing the hypothesis more fully. Clinically, you will be responsible for participant recruitment, assessment, injection with radioligand as needed on scanning days (after suitable training), maintaining records and ensuring compliance of the study with expectations of regulatory bodies and the sponsor and in drafting the final study report.
